Will depend on what alliance you ultimately want to travel with and times etc... but consider the following...
EY via AUH approx 27hrs
VA & EY & SA via MEL & AUH approx 30hrs
QF via SYD approx.17hrs
QF & SA via PER approx. 19.5hrs
SQ via SIN approx. 20 hrs
EK via DXB approx.26 hrs

QF via Sydney,
SQ via Singapore,
VA/SA via Perth.
These are the quickest options. QF/SQ have a better business class however the VA/SA combo is generally cheaper.

I would recomend CX via Hong Kong.
SQ flies their retro ftted B777-200s to JNB, whereas CX will be on their B77W from Hong Kong to JNB

CX is always good, however I think there may be a long layover in HKG. That said, If you want to spend the day in Hong kong between flights it may well suit.

This route has several combinations and tuzza1 is correct one is 37hrs effectivelly giving you a complete day layover in Hong Kong (available both ways) - the other combination (also available both ways) is 27hours making the transit in HKG approx 5.5hours. However, this route may not suit as well with your Velocity Platinum status.
