I been blessed with complimentary upgrade from business to first from LHR to SYD. However I traveling with my wife and she was not eligible for upgrade because we get ticket for her by QFF, so she will travel business on the same plane. So question - do they permit guests to Concorde Room in this circumstances?

Assuming you are flying BA LHR-SYD out of T5 then the very useful FlyerTalk BA Lounge Database (google it) says you are allowed one guest.
