I recently booked economy seat using cash to fly from Darwin to Perth and requested upgrade to business using points. I can see there are no business rewards seats, but business seats are available for cash. When can I expect Qantas to accept/decline my upgrade request? I thought domestic upgrades are confirmed straightaway as long as business seats are available.

According to their website it will be within 24 hours of scheduled flight departure.
Can be as early as 3 days before departure if you are Platinum.

That's not correct. Domestic upgrades are confirmed straightaway as long as business reward seats are available.
If no business reward seats are available, you'll have to wait to see if Qantas releases more, which might not be until the day of the flight.
If you don't hear back before the flight, your last resort is to request an "on departure upgrade" when you arrive at the airport, or at the lounge.
