Hi, I am flying out of Sydney Airport with Qantas to Shanghai in business class.

I am taking no checked in luggage, so I'll do an online check in. Do I get an express card for customs printed on my online boarding pass, or do I have to go to the Qantas desk and get an express card?

Go to the Qantas desk to get an express card for immigration. You will need to present at the Qantas desk anyway since agents will need to validate your documents and verify your ability to enter China on existing travel documents.

Can I do the above at Melbourne? Or it has to be at Sydney?

You can do it in MEL. If they forget your express card just remind them.
