I understand to upgrade from premium economy to business class on Cathay Pacific you must be booked in either R or W. If I have an E class fare am I eligible for at the airport upgrades to business with points?

No E class is not available for upgrade even at the airport .You will need to upgrade the booking class to W or R however the cost difference with CX is usually disproportionate !
