I have booked PER to LHR on QF9. It now transpires that I will be in Melbourne the day before my PER/LHR flight. I would contemplate staying overnight in Melbourne if I could simply book MEL to PER on the same QF9 flight and then (using both tickets) check in at Melbourne for both QF9 sectors contemporaneously as if I were flying MEL / PER / LHR and only transitting PER. Any advice would be welcome.

If you book/booked both through qantas you shouldn’t have any problem connecting the two and checking bags all the way through.
